We're a couple cycling around Asia. We started in Bali and cycled through Java, Singapore, Malaysia and are in our last month of Thailand. We aim to enter Cambodia via O Smach.
I included our rough itinerary for our month in Cambodia (starting March).

Hi Nicholas, we have recently returned from a Ho Chi Minh to Siem Reap circuit, generally the opposite of what you are proposing. Generally the roads in Cambodia are worse and there are some less scenic parts as well as some smaller towns that aren't really inviting. The section from Siem Reap to Phnom Penh was noted as being one of those sections, so we used a local bus for that part. It was easy and they only charged $5 for bikes. We were also a bit more pressed for time, so wanted to spend more days at the temples in Seam Reap, rather than on a challenging section of their road system.
The section south from PP to Phum Prey Nhoek was a bit slow since we wanted to take the small village roads, but they all run east-west so we had a lot of zig-zagging to get there in one day. We also went south on 41, rather than highway 3 or 4 as we were advised it was a quieter route. It was straight and uneventful, but there wasn't a lot of towns along the way.
There is also a very quiet dirt road along most of the coast between Kampot and Kep. It is visible in google earth, but not clear on too many maps. We have it all on GPS, if you can't find it.
We also crossed the border at Ha Tien into Vietnam and did a side trip to Phu Quoc. The main part of the island Duog Dong and the south are good, but the trip north wasn't worth it. There were lots of construction vehicles, but not too much else to see.
Since you said you are going to Vietnam after, the following might be useful.... The ferry also takes you to Rach Gia, which was a very nice town and provides a good low traffic route through to Ho Chi Minh, via Can Tho, Vinh Long and Ben Tre.
You can also avoid the major roads (CT01) by coming from the south through Ben Luc and staying on the north side of the river between An Thanh and Tan Tuc and the service road gets you to the interchange (QL1) which then has a scooter lane through to the middle of HCM.
